| Description | Blank LNPs are nanoscale particles formed by self-assembly of multiple lipid components. Its main components include phospholipids, which can form a lipid bilayer structure similar to that of a cell membrane and provide the basic framework for LNPs. Cholesterol is often added to adjust the fluidity and stability of the lipid bilayer, allowing LNPs to maintain structural integrity in different environments. Some LNPs also contain some lipids, such as cationic lipids. The positive charge of cationic lipids can be combined with negatively charged nucleic acids (such as mRNA, siRNA) through electrostatic interactions, which facilitates the subsequent packaging and delivery of nucleic acid drugs. Although no drugs are encapsulated in blank LNPs, the drug-binding properties are retained. The entire LNP system has a relatively uniform particle size distribution and a large specific surface area, which allows it to have a high drug loading capacity and good encapsulation rate when used as a drug carrier. At the same time, its nanoscale size gives it the ability to pass through biological membranes and avoid phagocytosis by the monocyte-macrophage system, which is conducive to long-term circulation in the body and increases the chance of drug enrichment at the target site. |
