Introduction to Cell Magnetic Sorting Methods

Positive Selection

This method utilizes magnetic beads conjugated with antibodies specific to the surface of target cells (CD4, CD8, etc.) to bind directly to the target cells in the sample. Under the influence of a magnetic field, the target cells labeled with magnetic beads are adsorbed onto a magnetic rack or sorting column, while non-target cells are eluted and removed.

  • Directly captures target cells, making it suitable for isolating specific cell subsets.
  • Few steps; typically completed within 30 minutes.
  • Suitable for samples with low target cell content or when high-purity separation is required (e.g., selection of tumor-infiltrating lymphocytes).

Negative Selection

A mixture of magnetic beads conjugated with antibodies specific to non-target cells (e.g., targeting CD11b, CD45, or Ter119) is used to label and remove all non-target cells. Target cells remain unlabeled and are retained in the supernatant. After removing the magnetically labeled non-target cells using a magnetic field, pure target cells are obtained.

  • Target cells do not bind to any magnetic beads or antibodies, maintaining their original state, making them suitable for subsequent culture or functional experiments.
  • Avoids potential activation or damage to target cells caused by antibodies or magnetic beads.
  • Suitable for experiments with strict requirements regarding cell function.
