| Description | Based on sequence similarity to VEGF-A, a gene encoding a VEGF homolog was recently identified in the genome of the sheep oral herpes virus (Orf virus, OV) (Lyttle et al., 1994). Different strains of the sheep oral vesicular disease virus exhibit significant amino acid sequence similarity to VEGF-A and have been described as a viral virulence factor that appears to have originated from a captured host gene. All eight cysteine residues in the central cysteine motif, which is characteristic of VEGF family members, are as conserved in the VEGF-E protein as they are in other residues. Alignment of all mammalian VEGF sequences indicates that VEGF-E differs from previously described VEGFs but is most closely related to VEGF-A. Similar to VEGF-A, VEGF-E has been found to bind to VEGF receptor 2 (KDR) with high affinity, leading to receptor autophosphorylation. However, unlike VEGF-A, VEGF-E cannot bind to VEGF receptor 1 (Flt-1). Furthermore, VEGF-E cannot bind to VEGF receptor 3 (FLT-4) either. Therefore, VEGF-E is a potent angiogenic factor. |
