Description LNP@Fluc-mRNA is a gene delivery system that uses lipid nanoparticles to efficiently encapsulate the mRNA encoding firefly luciferase (Fluc). Fluc can catalyze the oxidation reaction of luciferin to produce bioluminescence. It is often used as a reporter gene to monitor gene expression, cell activity and molecular events in the body in biomedical research. However, naked mRNA is susceptible to nuclease attack and is difficult to effectively deliver to target cells and achieve stable expression. Lipid nanoparticles are composed of ionizable lipids, auxiliary lipids, cholesterol, and PEGylated lipids, and have good biocompatibility, modifiability, and nucleic acid encapsulation capabilities. It tightly wraps Fluc-mRNA, not only protecting the mRNA from degradation, but also accurately delivering the mRNA into target cells through fusion with the cell membrane or endocytosis. After entering the cell, Fluc-mRNA uses the intracellular translation mechanism to guide the synthesis of firefly luciferase, and then detects fluorescence signals to achieve real-time and sensitive monitoring of intracellular biological processes.
