| Description | LNP@Fe3O4/ICG is a multifunctional nanocomposite that combines magnetic and fluorescent properties. Fe3O4 nanoparticles and indocyanine green are co-encapsulated inside lipid nanoparticles (LNP). Lipid nanoparticles are mainly composed of ionizable cationic phospholipids, cholesterol, auxiliary phospholipids and polyethylene glycol modified phospholipids. Under physiological pH conditions, ionizable cationic phospholipids are neutral, protonated and positively charged in acidic environments such as endosomes, and can tightly combine with negatively charged or polar Fe3O4 and ICG through electrostatic interactions, hydrophobic interactions, etc. to achieve efficient encapsulation. Cholesterol enhances membrane fusion and helps LNP@Fe3O4/ICG to be taken up by cells and promote the escape of Fe3O4 and ICG from endosomes into the cytoplasm. Auxiliary phospholipids stabilize the phospholipid bilayer structure and maintain the stability of LNP morphology. Polyethylene glycol-modified phospholipids are located on the surface of LNP to prevent particle aggregation, prolong their circulation time in the body, and avoid rapid clearance by the immune system. Iron tetroxide has superparamagnetic properties and can achieve directional movement and enrichment under the action of an external magnetic field; indocyanine green is a near-infrared fluorescent dye that can produce fluorescence signals when excited by light of a specific wavelength. After the two are combined with LNP, they not only improve the stability, but also endow the complex with multiple functions such as targeted delivery, magnetic manipulation, and fluorescent tracing. |
