| Description | DOTAP-DOPE Cationic LNP is a representative cationic liposome system used in the field of non-virus gene delivery. It is an optimized compound of the cationic lipid DOTAP (providing permanent positive charge) and the neutral auxiliary phospholipid DOPE (usually at a 1:1 molar ratio). DOTAP combines and compresses negatively charged nucleic acids (DNA, siRNA, mRNA, etc.) through the strong positive charge generated by its quaternary ammonium salt head group to form a stable nanocomplex; while DOPE relies on its unique conical molecular structure to promote the fusion or destruction of liposomes and endosomal membranes after endocytosis, enhance endosomal escape efficiency, and effectively release nucleic acids into the cytoplasm. |
