Description DMPC-chol-DSPE-mPEG2000 (50-45-5, mol-mol)lipid nanoparticles are functionalized lipid nanoparticles with a low PEG modification ratio (5 mol%), using DMPC-chol-DSPE-mPEG2000 (50: 45: 5)ternary compound formula. This system uses high-fluidity DMPC (phase transition temperature ~23°C)as the main phospholipid, combined with a high proportion of cholesterol (45 mol%)to enhance membrane stability, and introduces low-content DSPE-mPEG2000 (5 mol%)to achieve mild PEGylation on the surface. This design retains the temperature-sensitive characteristics of the DMPC system, while reducing plasma protein adsorption through the steric hindrance of the PEG layer and extending the body circulation time.
