Description Cho-es-Lys (cholesterol-succinate-lysine conjugate) is a designer amphipathic molecule that combines the membrane-anchoring ability of cholesterol with the charge and reactivity of amino acids. The molecule covalently connects the hydrophobic cholesterol steroid ring structure to the hydrophilic lysine residue through a succinate linking arm to create a structurally multifunctional lipid derivative. The cholesterol moiety serves as a membrane-anchoring module that is able to firmly insert into the hydrophobic core of lipid bilayer membranes, liposomes, or nanoparticles through hydrophobic interactions, giving the entire molecule a stable positioning in the biological membrane environment. Not only does the succinic acid linker provide suitable chain length and flexibility, its carboxylic acid group (usually present as an amide bond after being attached to lysine) also increases the polarity of the molecule. The terminal lysine residue is the functional core of the molecule: its side chain carries a net positive charge (ε-amino protonation) under physiological pH conditions, which makes the entire Cho-es-Lys molecule exhibit cationic properties.
